Withaferin-A (Wi-A) exhibits approximately 90% cytotoxicity against normal cells at 1 μM, while 3β-Methoxy-2,3-dihydrowithaferin A (3βmWi-A) remains well tolerated even at 5-10 μM . Wi-A inhibits clonogenic formation at concentrations as low as 0.1 to 0.3 μM, whereas 3βmWi-A shows no significant effect even at 10-fold higher concentrations . Wi-A treatment increases ROS levels in cells, while 3βmWi-A (0.3, 0.6, or 1.5 μM) reduces ROS levels after cell treatment .
