Depletion of CD8+ T-cells in mice using rat or mouse version of anti-CD8 antibody clone YTS 169.4. Mouse IgG2a antibody format of anti-CD8 clone YTS 169.4 (purple, dashed line) and Rat IgG2b of the same clone (orb613788; blue, dashed line) were used to deplete CD8+ T cells in a mouse model. Only species-matched mouse version managed to deplete CD8+ cells over a prolonged period of time (over 40 days). Continuous lines show appropriate isotype controls.

Flow-cytometry using anti-CD8alpha/Lyt-2 antibody YTS 169.4 Mouse lymphocytes were stained with an anti-CD44 control (orb613885, panel A) or the rabbit-chimeric version of YTS 169.4 (orb613787, panel B) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a AF488 conjugated donkey anti-rabbit antibody and cells analysed on a FACSCanto flow-cytometer.

Flow-cytometry using the anti-CD8 alpha/Lyt-2 antibody YTS 169.4 Mouse thymocytes were stained with unimmunized rabbit IgG antibody (black line) or the rabbit-chimeric version of YTS 169.4 (orb613787, blue line) at a concentration of 10 µg/ml for 30 mins at RT. After washing, bound antibody was detected using anti-rabbit IgG JK (FITC-conjugate) antibody (129936) at 2 µg/ml and cells analyzed on a FACSCanto flow-cytometer.
