Plant/fungal DNA extraction kit

This kit extracts total DNA from 100–500 mg of fresh plant tissue, 20–50 mg of dried plant tissue, or 100–300 mg of fungi. Frozen in liquid nitrogen, samples are homogenized to disrupt cell walls before lysis buffer is added to liberate genomic DNA. Chloroform extraction removes interfering proteins and polysaccharides, after which the aqueous phase is loaded onto a spin column. DNA binds to the column membrane as remaining proteins and inhibitors are washed away with Buffers RPB and WB. Finally, high-purity DNA is eluted using Buffer TE for direct use in downstream molecular assays.
